Prions play powerful role in the survival and evolution of wild yeast strains

Wednesday, February 15, 2012

Prions, the much-maligned proteins most commonly known for causing "mad cow" disease, are commonly used in yeast to produce beneficial traits in the wild. Moreover, such traits can be passed on to subsequent generations and eventually become "hard-wired" into the genome, contributing to evolutionary change.

Prions were first found to produce heritable new traits more than a decade ago in laboratory studies of simple baker's yeast. The key discovery then was that some proteins could spontaneously switch from a normal shape into a self-perpetuating prion conformation. The switch to the prion state alters protein function, which can result in the appearance of new traits, some helpful, some detrimental. Sophisticated cellular machinery ensures that replicating prion templates are chopped into pieces that can be passed to daughter cells during cell division. Importantly, the rate at which proteins switch into and out of the prion state increases in response to environmental stress, suggesting that they are part of an inherent survival mechanism that helps yeasts adapt to changes in their surroundings.

Yet, as compelling as the case for this protein-based mechanism of inheritance is, its biological significance has been hotly debated for one key reason: prions capable of modifying phenotypes have never been found in nature. Until now.

In a massive undertaking, Whitehead Institute scientists have tested nearly 700 wild yeast strains isolated from diverse environments for the presence of known and unknown prion elements, finding them in one third of all strains. All the prions appear capable of creating diverse new traits, nearly half of which are beneficial. These unexpected findings, reported in this week's edition of the journal Nature, stand as strong evidence against the common argument that prions are merely yeast "diseases" or rare artifacts of laboratory culture.

"A huge amount of effort has gone into studying this paradigm-shifting mode of inheritance, but with no real understanding of whether it's genuinely important biologically," says Daniel Jarosz, co-first author of the Nature paper and a postdoctoral researcher in the lab of Whitehead Member Susan Lindquist. "Now it seems clear they do influence the way natural yeasts cope with changing environments and evolve in response to stress."

The hunt for prions in wild yeast strains began in the Lindquist lab when Jarosz and Randal Halfmann, then an MIT graduate student and now a fellow at the University of Texas Southwestern Medical Center, gathered hundreds of wild strains from stock centers all over the world. They then conducted a chemical screen for one well-studied prion, [PSI+], and found it in 10 wild strains. Genetic manipulations confirmed its status as a true prion. They then observed the effects of [PSI+] on biological traits by eliminating the prion conformation in these strains and exposing them to natural stresses, such as high acidity and the presence of antifungal agents. In one strain isolated from Beaujolais wine, for example, the prion resulted in the emergence of traits that could be beneficial or detrimental, depending upon environmental conditions. Another well-known prion, [MOT3+] was found in six wild strains.

To determine whether the wild yeasts might harbor other unknown prion elements, Jarosz and Halfmann exposed sets of cultures of all the wild strains to the same chemical protocol that switched [PSI+] and [MOT3+] out of their prion states. In all, 255 strains demonstrated different phenotypes under varying stressors after this treatment.

"We certainly didn't expect to see this much prion-based phenotypic diversity," Jarosz says. "It's remarkable."

Another surprise was that approximately 40% of the traits produced by the wild prions proved to beneficial to growth in the dozen different environmental conditions tested.

"How frequently beneficial they are suggests that the prions have already been subject to previous, positive selective events," says Lindquist. "We see them as part of a bet-hedging strategy that allows the yeast to alter their biological properties quickly when their environments turn unfavorable."

Convinced of the impact prions have had on yeast evolution, Lindquist speculates that these shape-shifting proteins may be "remnants of early life," from a time when inheritance was predominantly protein-based rather than nucleic-acid based. She also theorizes that prions may play such roles beyond yeast, and her lab intends to take similar approaches in the hunt for prion activity in other organisms.

Deadlock on Syria: Likely crimes against humanity, but no plan of action

Turkey's foreign minister discussed Syria for hours with US officials Monday, but no 'road map' for action resulted, even as the top UN human rights official warned of crimes against humanity.

The UN?s top human rights official declared Monday in New York that international inaction on the intensifying crisis in Syria likely is enabling the regime to commit crimes against humanity.

But in Washington, senior officials of the United States and Turkey, two countries that have called publicly for President Bashar al-Assad to cede power, seemed incapable of making any progress toward developing a plan of action, humanitarian or otherwise.

Secretary of State Hillary Rodham Clinton hosted Turkish Foreign Minister Ahmet Davutoglu, who also met with Defense Secretary Leon Panetta and White House national security adviser Thomas Donilon.

Secretary Clinton announced she will be attending a ?friends of the people of Syria? meeting in Tunis Feb. 24. That meeting, hosted by the Arab League, is expected to address ways of boosting humanitarian assistance to Syria?s besieged civilian population. The Arab League is also proposing an even more problematic UN peacekeeping mission for Syria.

But several hours of meetings did not appear to produce the ?road map? for the international community?s involvement in Syria ? particularly in addressing a mounting humanitarian crisis ? that Mr. Davutoglu had spoken of at the outset of his Washington visit last week.

After the meetings, Clinton said the US would continue to work on tightening international sanctions on the Assad regime and will look for ways to increase delivery of humanitarian assistance in the 10 days before the ?friends of Syria? meeting.

But she offered little hope that the Arab League?s proposal for a peacekeeping force in Syria could attain the Security Council support it would require. ?There are a lot of challenges to be discussed as to how to put into effect all of [the Arab League?s] recommendations, and certainly the peacekeeping request is one that will take agreement and consensus,? she said.

Apple asks labor group to probe China suppliers

Workers are seen inside a Foxconn factory in the township of Longhua in the southern Guangdong province in this May 26, 2010 file photo. The Fair Labor Association has begun audits of Apple suppliers' labor practices in China, at the company's request.

By Eve Tahmincioglu

Apple announced Monday a non-profit labor group has started examining working conditions at some of its suppliers in China, as the company tries to calm a growing storm over how the workers who build iPads and iPhones are treated.

One particular supplier, Foxconn, has come under fire for working conditions at its facilities including alleged?child labor violations and unsafe work environments. Labor advocates have long noted the problems, but the working conditions?got more exposure following a scathing expose in the New York Times last month. The Foxconn plants?have seen a rash of suicides in the past year.

Protests have swelled over Apple's labor issues, including demonstrations at the company's stores around the globe. There's also a petition?on Change.org?titled "Apple: Protect Workers Making iPhones in Chinese Factories" which has more than 200,000 signatures. The petition was started by Mark Shields, an Apple customer who appealed to the company to: "Please make these changes immediately, so that each of us can once again hold our heads high and say, 'I?m a Mac person.'"

Last week, Foxconn's computers were hacked as part of the growing protests, according to The Guardian.

The bad press has been rare for Apple, whose stock hit $500 a share for the first time ever Monday,?so it?s not unexpected that the company would move to take some action. It?s unclear, however, whether these audits will lead to change.

The Fair Labor Association (FLA) will conduct what Apple calls, ?special voluntary audits? of Foxconn?s plants in Shenzhen and Chengdu, China. The first inspection commenced Monday, according to a company statement. The findings will be posted on the organization?s website in March. www.fairlabor.org.

?We believe that workers everywhere have the right to a safe and fair work environment, which is why we?ve asked the FLA to independently assess the performance of our largest suppliers,? said Tim Cook, Apple?s CEO. ?The inspections now underway are unprecedented in the electronics industry, both in scale and scope, and we appreciate the FLA agreeing to take the unusual step of identifying the factories in their reports.?

Some labor advocates aren't as hopeful.

"The entry of Apple to FLA is a welcome development," said Mary Gallagher, director of the University of Michigan's Center for Chinese Studies, and associate professor for political science. "However, I'm not optimistic that conditions in Apple's supplier factories will change if we only rely on occasional inspections from an overseas group."

Chinese workers, she continued, "are increasingly aware of their legal rights at the workplace but they often lack the proper tools to realize enforcement of those rights, such as the right to strike and the right to organize collectively."

Li Qiang, director of China Labor Watch based in New York, said "FLA speaks on behalf of the companies, not workers.?If Apple wants the inspections be accurate and trustworthy, it should have labor advocacy groups who speak for the workers involved in the inspection process."

A call to FLA officials was not immediately returned.

The FLA will interview thousands of workers at the facilities, according to Apple, and review?compensation, safety, and even?the dormitories where Foxconn houses its employees.

The company's own audits of its final assembly plants, conducted since 2006, have done?little to alter conditions at the facilities, according to labor experts and Apple?s own report on the state of its Chinese manufacturing partners, which was?released last month.

China Labor Watch's Qiang said he sees Apple's move as more of an attempt to rebuild its public image rather than?to help workers.

"What Apple should do now is to take action to solve the problems and improve the labor conditions in their supplier factories, not to conduct inspections and put the factories into the media and public's attention," he maintained.?

Easing Wrist Joint Pain

Published by Sherry | February 10, 2012 | | 86

One of the more potent treatments for wrist joint pain caused by inflammation is injections. The injections contain an anesthetic and steroid. It can be administered in different dosages. This means the injection can be milder or stronger. Whatever the strength of the injection it will last a lot longer than the average anti inflammatory. A single injection can bring relief that lasts for a few weeks or months depending on the strength of the dose. An injection is a very good means of getting rid of pain and inflammation for long periods but there is the risk of the tendons become weak. An injection can also noticeably lighten the skin.

Before a doctor prescribes an injection the patient will usually take anti inflammatory medication. Many people use this medicine and also go for alternative treatments such as massage, acupuncture and herbs. A lot of people say the alternative treatment does bring some relief and it also encourages them to relax.

This is good because pain causes stress.

The wrist is what connects the hand to the arm. When the wrist is in pain it makes the use of the hand difficult. It can be so painful that a sufferer will opt for surgery if none of the other alternatives work. The surgery consists of the joint being replaced. This is a treatment that has a high success rate because it lasts long term. Medical doctors perform surgery as a last resort.

After this kind of surgery you will need to visit a physiotherapist to learn how to use your hand with the replacement joint. The wrist joint pain and inflammation will be gone and the vast majority of patients have found it well worth the effort. However, it is important to bear in mind that over time the synthetic replacement will deteriorate and may need to be replaced.

James K. Falcon is a wrist joint pain expert and the director of JointPainTreatmentInfo.com. James K. Falcon provides honest information and advice on things like joint pain management and much more. Check out the website today!
